I've recently got my new EE sim after transferring from another network.
I registered my Top-Up card by texting ETU [card number] to 150, thinking that it is registered to the sim/account rather than the phone number. After this I requested to transfer my old number to EE.
Will I be able to use my top up card as normal after this, or will I need to go and get a new card to register with my ported number?

Try sending the ETU to 150 again & see what it responds.

I got my number transferred and tried sending ETU. Came back as registered, brilliant 😀
