Hello!
I tried to replace my ESIM many times online, however, I cannot transfer it to my replacement IPhone.
I do not have my old phone, I have no service or use of my phone number which is essential to me.
My sister owns the contract, I cannot visit in store and she does not have time to either.
Before she orders a replacement physical Sim online,
is there a way I can buy a new EE sim from a supermarket and she contacts to link this new sim with my current contract through online/telephone?

The short answer is no. Any new EE SIM you buy from a supermarket will be pre-linked to a PAYG account.
