Not on the Samsung s6.
You CAN actually get it on any device right now. You need to set your device language to English US. Then you must go to the Google app on the Google play store. Scroll to the bottom and sign up to the developer version. Wait a while for it to update and you'll have the Google assistant. However, be warned. The US version didn't understand my English accent of "Ok Google" until I did my best impression of an America... Also, the moment you turn you language back to normal it'll "update" to "downgrade" the app.

When you say not on the S6... Does that mean it's NOT going to get it? I thought it was rolling out to all devices running Marshmallow or higher.

I didn't mean that, but it's come out anyways. Got it around 4 days ago. 12th April update enabled Google assistant for me. It only appears when holding the home button, opposed to pressing voice search.
This is without changing my phone's language or being in the beta programme.

Yup got Google Assistant on my last update. Don't use it, will see how Bixby is when I get S8+

In theory when you long press the home button GA should ask how can I help you, however I still get GoogleNow

Have you ever used the Edge Sense yet.
It seems the current app it opens is Google Assistant.
Just squeeze the edge of the phone. If this is not set up then go to settings to set up.

I have indeed but that just opens google now. Checked ALL settings, nowhere can it be found, only google now. Very odd!
